Genetics Lab­ Lab Exercise 1  8/23/16    ● Metric system is based on the decimal system  ○ Have to know femto to peta for the class    ● Three advantages to scientific notation  ○ Allows you to work in powers of 10,100, 1000  ○ Easy to report and determine # of sig figs in a measurement   ○ Allows us to do multiplication and division of large numbers w/o a calculator  ■ Multiple coefficients, add exponents    ● Micropipettes are used to measure small amounts of liquid   ○ Adjustable, have different ranges of volume  ■ P1000­ Max 1 mL (1000uL), min 100uL  ■ P200­ Max 200uL, min 20uL  ■ P20­ Max 20uL, min 2uL  ■ P2­ Max 2uL, min 0.1uL    ● Serial dilutions­ well­quantified dilutions, take a very high concentration of cells or  reagent and dilute it down to a manageable concentration  ○ Used for microbe pipetting or for determining unknown concentrations  ○ Concentration dilutions help make a Standard Curve or Beer’s Law curve, then  helps find out concentration of unknown    ● How to find molarity:  ○ 0.5% is 0.5g/100mL or 5g/1000mL  ■ (5g/1000mL) x (1/319.85g)= 1.56x 10^­6  ● In this scenario, 319.85g is the molar mass     ● How to find dilution:  ○ Dilution = amount brought over/ total volume  ○ Find a standards concentration by multiplying the starting molarity and the  dilution factor of that particular standard    ● Sterile technique­ procedures where cultures can be manipulated w/o infecting the  student or contaminating the cultures/ lab  ○ Use sterile pipets and pipetmen  ○ When using spreader, the EtOH is what sterilizes it  ■ Flame is used to burn it off    ● Beer’s Law  ○ A= E x l x c  ■ A is absorbance  ■ E is molar absorptivity  ■ L is pathlength of cuvette  ■ C is concentration in mol/L or molarity
