BIOL 5600__________________________________________________________________ __________3-1-16 Sympathetic Nervous System Presynaptic effects 1) Block neurotransmitter synthesis -Ex. of drug that does this: >Demser -Blocks synthesis of norepinephrine >In fault, blocking epinephrine synthesis (NE is precursor to E) -By blocking enzyme tyrosine hydroxylase >In fault, blocking synthesis aldopa (precursor to NE) -Decrease in sympathetic effect -Once used to treat hypertension, but too wide range of effect *Important thing about drugs… how specific are they 2) False neurotransmitter -Precursor molecule called methyldopa (aldomet) >Substitute for aldopa -Gets converted into alpha methyl norepinephrine -Does not bind alpha 1 or beta 1 receptors -It does bind alpha 2 receptors -Inhibitory -Decreases sympathetic activity -Alpha-methyl-NorE o Effector -Used to treat hypertension 3) Block release of NT’s -1 smelin -Decrease of NE/E secretion -Anti-sympathetic -Hypertension treatment 4) Block transport of NE back into the synaptic vesicles -Serpasil (reserpine) -MAOs are in cytoplasm of NMJ -When the transport or NE is blocked, the MAOs breakdown -So less NE leads to a down regulation of sympathetic effect 5) False displacement -Does not interfere with bindng directly >Has indirect effects -Makes NE more available and more active -Amphetamines (Controversy over whether there is binding to receptors, or just enhancing effect of NE) Ex. Dexatrine, increases sym. Effect -Treats hypotension, narcolepsy -Lots of side effects *All above is presynaptic! *Now talking about at the receptor level Adrenomimetics -Agonists at the receptors -Some are less specific than others A) Alpha 1 agonists -Promote sympathetic effects -Epinephrine/adrenalin -Epipen >High dose causes binds alpha and betas >Hypotension (shock) (anaphylactic shock) (mass vasodilation) -NE (levophed) >Alpha-receptors, and in high doses, B receptors >Pseudoephedrine -Basic OTC nasal decongestants -Constrict >Alpha 1 agonists B) Alpha 2 agonists 1) Clonidine (catapres) -Alpha 2 agonist -Promotes blood flow to coronary arteries -Causes inhibitory effect of vasodilation -Can decreases hypertension (treatment of hypertension)
