Affects of Ph3 Ph5 and Ph7 on the Benzoquinone reaction rate at twenty second intervals in a one hundred and twenty second time frame Amanda Doyle Section 6 Yuna Seo Introduction In this experiment treatments of Ph3 Ph5and Ph7 are added to culture tubes containing catechol and potato enzyme extract to determine the impact of different Ph levels on the oxidation of catechol Three trials are conducted for each treatment In twenty second intervals absorbance ODU is recorded and concentration mM is calculated for a period of one hundred and twenty seconds It is hypothesized that when the hydrogen ion concentration changes it will not affect the reaction time of catechol and stand no affect on the benzoquinone reaction rateAltematively if the hydrogen ion concentration changes it will affect the reaction time of catechol and the benzoquinone reaction rate Methods For Ph3 a culture tube was prepared containing 3ml of the buffer Ph3 and 5ml of 005M catechol The same concentrations were used to prepare culture tubes of Ph5 and Ph7The blank tube contained 8ml of buffer and lml of potato enzyme extract lml of potato enzyme extract was then added to each treatment tube The spectrophotometer was set to a wavelength of 500 and transmittance to 0 The blank was inserted and transmittance set to 100 The blank was removed and the sample with Ph3 was inserted The mode was then changed to absorbance Absorbance was recorded starting at zero and then every twenty seconds for one hundred and twenty seconds This process was repeated three times The same method was used for Ph5 and Ph7 After all absorbencies were recorded the concentrations for the three different buffers were calculated Results The level of buffer affected the concentration of benzoquinone over the one hundred and twenty second time period As the Ph became more basic ex From Ph3 to Ph5 the benzoquinone concentration increased over the given time The results of Ph3 and Ph5 reject the null hypothesis The results of Ph3 and Ph5 accept the alternative hypothesis Ph7 had unexpeted results and should be further tested Benzoquinone Concentration vs Time at pH 357 O Seriesl I SeriesZ A Series3 Linear Seriesl Concentration mM Linear SeriesZ Linear Series3 Time sec Note Ph3blue Ph5red Ph7green Conclusion The null hypothesis statesthat if the hydrogen ion concentration changes it will not affect the reaction time of catechol and stand no affect on the benzoquinone reaction rate This hypothesis was rejected and the alternative was accepted The results show changes in the concentration of Benzoquinone over time at the three different Ph levels Changes in Ph can affect the shape of an enzyme Enzymes are very speci c so changing the shape would change its function or recognition ability Errors could have been made in this experiment by not shaking test tubes adding the enzyme to early or miscalculated raw data This experiment should be repeated in order to compare results and determine how strong or if the data and results hold true
