Regulation of Gene Expression  Organisms only express genes when it is necessary o Transcription factors can neglect to appear  In prokaryotes, genes are found in operons (groups of genes transcribed at the same time to produce one mRNA  Lac Operon (lactose) produces a certain protein for each gene: LacZ= β­galactosidase (breaks down lactose), LacY= permease (imports lactose into cell), LacA= transacetytase (modifies lactose for use)  LacI codes for a repressor protein which stops transcription when it is unnecessary (when there is no lactose or when glucose can be used instead) o Binds to operator and blocks polymerase from attaching to DNA molecule o Lactose can also bind to the repressor which changes its shape so that it cannot bind to the operator  Promotor= CAP binding + KNAP binding (includes TATA box) + operator o TATA box can be both weak and strong  Strong: TATAAAA  Weak: TCTAAGA  When TATA box is weak, polymerase will fall off about 85% of the time  Only 15% expression o CAP can bind polymerase so that it does not fall off (for 100% expression)  Only bound by the small molecule cAMP which is present when glucose levels are low  Expressions: o No lactose at all: 0% expression o Lactose present:  15% when glucose is also present  100% when there is no glucose o No cAMP means glucose is present o W
