Challenges in Studying Mitochondrial Respiratory Complexes
Chapter 20, Problem 30(choose chapter or problem)
Questions & Answers
QUESTION: What would be some of the challenges involved in removing respiratory complexes from the inner mitochondrial membrane in order to study their properties?
ANSWER:Step 1 of 2
The respiratory complexes are made up of various proteins that are embedded in the inner mitochondrial membrane. Some of these proteins are tightly bound to the membrane, while others, like cytochrome c, are loosely attached.
There are several challenges when trying to remove these proteins from the membrane for study. The first is that proteins can easily change their shape and lose their function, a process known as denaturation. Hence, protein extraction needs more care and specialized reagents in comparison to DNA extraction.
